The two-time champion Sunrisers Eastern Cape will play the Paarl Royals in the second qualifying round. The Sunrisers Eastern Cape team finished the first round as the third-best team of the ILT20 2025 season after winning five of their ten games.  In addition, the Sunrisers advanced to the second qualifying after defeating the Johannesburg Super Kings in the Eliminator.
